ATM Mutations in Lymphoma.  We are collaborating with Joe Hacia, Ph.D. (USC), and have described the gene expression profiles of ATM mutations in mantle cell lymphoma utilizing an Affymetrix-based microarray system.  We plan to investigate the cause for the elevated ATM mutations and the effect in lymphoma development.

In 2006, we will begin using an ATM knockout mouse model to study the effect of environmental agents on the generation of abnormal hybrid T-cell receptor gamma-beta gene rearrangements.  

Gene Expression Analysis in Post-transplant Lymphoproliferative Disorders (PTLD).  In collaboration with Dr. Lou Staudt at the NCI, we have examined gene expression utilizing a cDNA microarray (Lymphochip) in PTLD.  We have studied 34 cases from this institution, and 10 cases from the University of Minnesota.  In addition, we have performed comparative genomic hybridization analysis on tumor DNA.  Comparative analyses will be performed between these disorders, infectious mononucleosis caused by Epstein Barr virus, and de novo subtypes of diffuse large cell lymphoma. 

Correlation of Gene Expression Array Data and Mutation Status in Diffuse Large Cell, Mantle Cell and Follicular Lymphoma.  In collaboration with Dr. John Chan, we are examining the gene expression patterns in the five major subtypes of lymphoma under the NCI Director’s Challenge “Towards a Molecular Classification of Tumors”.  Our laboratories are analyzing the p53 and bcl-6 mutation status in diffuse large cell lymphoma, and have published on ATM and p53 in mantle cell lymphoma to correlate with mRNA gene expression.  We aim to identify new targets or patterns of gene expression associated with mutation status.

Mutation screening – we utilize both denaturing grading gel electrophoresis and the Transgenomic WAVE DNA fragment analysis system. Opticon real time PCR instrument and laser capture microdissection are also available.
