PHAGE DISPLAY OF PEPTIDES TO ENHANCE THE IMMUNE RESPONSE TO THE PEPTIDE
Phage display of a peptide library or a single peptide on the filamentous phage, M13 have resulted in the amplification of the immune response to peptides in mice.
M13KE is a simple M13 derivative in which cloning sites have been introduced at the 5' end of gene III for display of short peptide sequences as N-terminal pIII coat protein fusions. Because this is a phage, all 5 copies of the pIII on the surface of each virion will be fused to the peptide.
We will design the oligonucleotide for the peptide, ligate it to the coat protein sequence and place the sequence into the M13KE vector. We will transform the E. coli, grow the phage to high titer, purify and concentrate using PEG,and quantitate virions. At this point, either the investigator or we can immunize mice used in hybridoma developement. The selection procedure, and the subsequent expression of single clones, will be performed by the investigator.
