Use one Request Sheet for Each Primer!
HOW TO FILL OUT A DNA SEQUENCING REQUEST FORM
1. For 8 or more samples with the same primer ask the Core Lab for a multiplesample sheet form (new prices). Otherwise, use one request sheet per primer. 2. Complete all requested information at the top part of the form. Account #’s are required before any work will be done. For bookkeeping purposes each form must be completed in its entirety. 3. Place an X in the box for the Double Stranded or PCR product and fill out corresponding blank lines with requested information. A. We are now requiring labs to measure their own Double Stranded plasmid samples using our fluorometer. The fluorometer is setup in Room 6001 and the protocol is located there. B. We cannot measure PCR products via fluorometry so a gel picture of the POST-PURIFIED PCR product is required. Inciate the sample volume loaded, a size marker and amount loaded. Tape the gel picture to the front part of the sheet. 4. Place an x in the box for Primer Selection. Remember ONE PRIMER PER SHEET! Standard Primers are listed below: Please fill in the primer selection M13-21For T3 Glprimer1 T7 terminator PGEX3' M13Reverse T7 Rvprimer3 T7eevpromotor PGEX5' M13-40For pucM13 Polyhedrin For KS SP6 pucM13Rev Polyhedrin Rev SK For Custom primers limit name to 5 characters and provide at NO EXCEPTIONS! HOW TO LABEL YOUR TUBES:Please provide DNA samples in 1.5 ml tubes and primers in 0.5ml tubes. The Core Lab will provide them if necessary.
