Oligonucleotide synthesis is performed using a BioAutomation Corporation MerMade that is capable of making 192 (2-96 well plates) oligos at one  time and two Perkin-Elmer Model 394 DNA/RNA Synthesizers.  A variety of product scales are available, ranging from 40 nanomoles to 25 micromoles.  RNA as well as DNA and phosphorothioate (sulfur) oligonucleotide synthesis is possible.  The DNA Synthesis lab also works with investigators who generate specialty amidites that are then coupled to make oligonucleotides using the automated synthesizers.

Several methods of purification of oligonuleotides are offered.  The Core Lab recommends specific purifications based on the type of chemistry used, product scale and length (number of bases) of the oligonucleotide product.  Purification methods available are desalting, cartridge, HPLC and PAGE.  Turnaround time is generally 48 hours or less for desalting and cartridge of smaller scale orders; one week for orders using HPLC or PAGE purifications.  Prices are very competitive with outside vendors.  The Core Lab offers complimentary primer design using Oligo Design Software that analyzes hairpin and dimer formation and provides information such as melting temperatures helpful in the set-up of PCR experiments.  After the primers are designed they are then made in house.
