Ettan Difference Gel Electrophoresis

Ettan DIGE Analysis*

The MSPCF offers consultation for a Ettan DIGE or difference gel electrophoresis. Ettan DIGE is a method of labeling proteins with an internal standard prior to 2D electrophoresis which results in both qualitative and quantitative data. By using an internal standard on every 2D gel, protein patterns can be matched across numerous gels, eliminating variation between them.

The whole system includes DIGE dyes, Typhoon 9400 scanning, DeCyder image analysis equipment. Equipment can be borrowed through the MSPCF from other investigators regularly performing 2D DIGE. DeCyder image analysis software is not available through the core but analysis is offered through Ludesi, a company specializing in 2D gel analysis. Their software can be downloaded for free at the following website: Ludesi *. Once a gel image is uploaded, Ludesi will analyse the image for a fee and produces results within a few days.

Ludesi image analysis can also be uploaded into a spot-picker which can be rented from the MSPCF ($200 per hour/ minimum of 1 hour). After the protein spots are picked, a simple protein digest can be performed, followed by analysis on one of our mass spectrometers.
