Amino Acid Analysis

Seventeen (17) amino acids are determined.  Other amino acids can be quantitated upon request.   Samples can be either liquid or blotted onto PVDF membrane. We perform vapor hydrolysis using 6N HCl with phenol and sodium sulfite added. 20-24 hours of hydrolysis under argon is generally used. Amino acids are then analyzed with a Hitachi L8800. The Hitachi L8800 uses an ion exchange column to first separate the amino acids. Color is developed (post-column-derivatization) with ninhydrin. This method may take samples which are contaminated with salts, urea, and small amounts of detergents. A sample of between 0.5 micrograms to 10 micrograms is necessary for analysis.
