{"id":131150,"date":"2026-04-08T10:30:00","date_gmt":"2026-04-08T15:30:00","guid":{"rendered":"https:\/\/www.unmc.edu\/newsroom\/?p=131150"},"modified":"2026-04-09T10:14:13","modified_gmt":"2026-04-09T15:14:13","slug":"chronic-myeloid-leukemia","status":"publish","type":"post","link":"https:\/\/www.unmc.edu\/newsroom\/2026\/04\/08\/chronic-myeloid-leukemia\/","title":{"rendered":"Fellow exploring gene rearrangement in chronic myeloid leukemia"},"content":{"rendered":"<div class=\"panel body-content\"><div class=\"panel__container\">\n<p>A pathology fellow is helping validate a FISH probe to test for a partner in an immunoglobulin gene rearrangement in chronic myeloid leukemia (CML).<\/p>\n\n\n\n<p>Phassawan Rungsiprakarn, MD, who started her two-year Laboratory Genetics and Genomics fellowship in July, received Research Grant Award funding from UNMC\u2019s Department of Pathology, Microbiology and Immunology for the project. She said the Human Genetics Laboratory at UNMC, headed by Dr. Zhenya Tang, fellowship director and her faculty mentor for the project, identified four CML cases with co-existing t(9;14)(q33;q32)\/IGH rearrangement (IGHr+) alongside t(9;22)(q34;q11.2)\/BCR::ABL1 fusion aberration. Two of them progressed from chronic phase to lymphoid blast phase, suggesting that IGHr+ may serve as a potential risk factor for this transformation. The LIM Homeobox 2 (LHX2) gene has been postulated as a potential partner for IGHr+ in these cases.<\/p>\n\n\n\n<p>Dr. Rungsiprakarn said they have developed a customized IGH::LHX2 dual-fusion Fluorescence in Situ Hybridization (FISH) probe set in house. Currently, there is no commercially available ready-to-use LHX2 FISH probe. While custom LHX2 FISH probes can be ordered from commercial vendors, they are expensive and difficult to optimize.<\/p>\n\n\n\n<p>They used the customized probe to determine the presence and status of IGH::LHX2 rearrangement in the pre-identified CML cases. In addition, they hope to develop a reverse transcription polymerase chain reaction method for semi-quantitative evaluation of LHX2 expression and determine the LHX2 mRNA levels in the study cases. They also will evaluate LHX2 expression with immunohistochemistry and expand this strategy to investigate other CML cases in their databank that may harbor potential t(2;9)\/IGK::LHX2 rearrangement and t(9;22)\/IGL::LHX2 rearrangement, if available.<\/p>\n\n\n\n<p>The RGA program was created by the department in 2024 to provide up to $10,000 in funding for high-quality research projects leading to publications by departmental clinical trainees for whom other funding sources are not available. The aim of this program is to advance the experience and mentorship in clinical and translational research.<\/p>\n\n\n\n<p>Dr. Rungsiprakarn, who earned her MD at Khon Kaen University Faculty of Medicine in Thailand, said she\u2019s grateful to be at UNMC. \u201cI had heard that this place has a strong molecular diagnostic lab and hematology services. I feel this is the right place to learn about genetics. So I feel very fortunate to be here, to be the first fellow in LGG here at UNMC.<\/p>\n\n\n\n<p>\u201cI came to this country because I would like to work on something that has impact on a global scale,\u201d she said. \u201cWith research, your findings can help people on the other side of the world. As a doctor from abroad, I found genetics fulfilling because there\u2019s always something new.
