{"id":47648,"date":"2002-07-30T00:00:00","date_gmt":"2002-07-30T05:00:00","guid":{"rendered":"https:\/\/www.unmc.edu\/newsroom\/2002\/07\/30\/unmc-dentistry-researchers-receive-209000-grant-tostudy-role-of-protein-in-multiple-sclerosis\/"},"modified":"2022-08-29T10:47:27","modified_gmt":"2022-08-29T15:47:27","slug":"unmc-dentistry-researchers-receive-209000-grant-tostudy-role-of-protein-in-multiple-sclerosis","status":"publish","type":"post","link":"https:\/\/www.unmc.edu\/newsroom\/2002\/07\/30\/unmc-dentistry-researchers-receive-209000-grant-tostudy-role-of-protein-in-multiple-sclerosis\/","title":{"rendered":"UNMC dentistry researchers receive $209,000 grant to\n\nstudy role of protein in multiple sclerosis"},"content":{"rendered":"<div class=\"panel body-content\"><div class=\"panel__container\"><p>Thomas Petro, Ph.D, Department of Oral Biology, UNMC College of Dentistry,<\/p>\n<p>has been awarded a $209,000, three-year grant from the National Multiple<\/p>\n<p>Sclerosis Society to study a cell protein called Interleukin-12 (IL-12)<\/p>\n<p>to determine its role in multiple sclerosis.<\/p>\n<p>The societys President and Chief Executive Officer, Mike Dugan, said<\/p>\n<p>the research is a promising and difficult study in the interest of all<\/p>\n<p>those who are living with multiple sclerosis.<\/p>\n<p>Dr. Petro, who has been studying IL-12 for 10 years, and for the last<\/p>\n<p>three years as it applies to multiple sclerosis, said despite the known<\/p>\n<p>association between multiple sclerosis and the protein, there is a lack<\/p>\n<p>of knowledge about how it is stimulated during the disease.<\/p>\n<p>We want to know what happens in the first few hours in the cell following<\/p>\n<p>its infection by a virus that causes multiple sclerosis symptoms in a mouse<\/p>\n<p>model, said Dr. Petro, principal investigator of the grant.. Despite<\/p>\n<p>several treatments that have been widely accepted, MS therapies have not<\/p>\n<p>been developed which exploit the<\/p>\n<p>signaling mechanisms that lead to the expression of key genes associated<\/p>\n<p>with disease, Dr. Petro said.<\/p>\n<p>Dr. Petro said all humans produce IL-12, which is a small protein produced<\/p>\n<p>by macrophage cells that communicate immune system responses. In regulated<\/p>\n<p>amounts, IL-12 is important in preventing many diseases, including cancer<\/p>\n<p>and tuberculosis.<\/p>\n<p>The problem is when excessive amounts of IL-12 are produced. Dr. Petro<\/p>\n<p>hypothesizes that excess IL-12 is produced in the mouse brain following<\/p>\n<p>an infection by an MS-model virus.<\/p>\n<p>An excess of the protein in the brain in mouse models causes multiple<\/p>\n<p>sclerosis symptoms, similar to those in humans. He and his team hope to<\/p>\n<p>determine the details of the key signaling mechanism that leads to a MS-like<\/p>\n<p>disease in the mouse virus model.<\/p>\n<p>We expect that similar if not identical signaling mechanisms play a<\/p>\n<p>role in humans, Dr. Petro said. In the mouse, if IL-12 is blocked, then<\/p>\n<p>symptoms get better, but we have yet to test this in humans. This gap in<\/p>\n<p>knowledge is important to fill since IL-12 expression is a feasible target<\/p>\n<p>for MS therapy.<\/p>\n<p>We have all the tools available to come up with an answer to exaggerated<\/p>\n<p>production of IL-12. Of course, we realize sometimes things work great<\/p>\n<p>in the test tube but dont always work out in the clinic.<\/p>\n<p>The teams long-term goal is to inhibit or reduce the cellular activity<\/p>\n<p>of IL-12 and develop therapeutic strategies for disrupting the process<\/p>\n<p>so symptoms can be reduced.<\/p>\n<p>If you could somehow lower the IL-12 production, the thought is that<\/p>\n<p>this will help people by reducing or controlling their MS symptoms. Reducing<\/p>\n<p>the IL-12 activity seems to help disease symptoms in mice, Dr. Petro said.<\/p>\n<p>The way MS works is that you have an initial episode with symptoms.<\/p>\n<p>Then after some time you may get better. Later you may have a recurrence<\/p>\n<p>of symptoms and the symptoms may get worse. All the while you know the<\/p>\n<p>symptoms will come back, he said.<\/p>\n<p>Dr. Petro said he and his colleagues also will study in the laboratory,<\/p>\n<p>various effects of drugs on IL-12.<\/p>\n<p>If we can figure out with pharmaceuticals how to inhibit IL-12 gene<\/p>\n<p>transcription factors, then maybe were on to something, then it actually<\/p>\n<p>may be possible to do something at the IL-12 gene to reduce or stop symptoms<\/p>\n<p>of MS. This goal is within reach because presently, several novel anti-cancer<\/p>\n<p>therapies have emerged from basic research dealing with cell signaling<\/p>\n<p>mechanisms. It is now time to explore similar possibilities for treatment<\/p>\n<p>of MS.<\/p>\n<p>Multiple sclerosis is a chronic, often disabling, disease of the central<\/p>\n<p>nervous system. Symptoms may be mild, such as numbness in the limbs, or<\/p>\n<p>severeparalysis or loss of vision. There currently is no cure for the<\/p>\n<p>disease. Until recently, steroids were the principal medications for MS.<\/p>\n<p>While steroids cannot affect the course of MS over time, they can reduce<\/p>\n<p>the duration and severity of attacks in some patients.<\/p>\n<p>Although the disease is not fatal, it causes weakness, tremors, loss<\/p>\n<p>of vision, cognitive changes, depression and other problems. About half<\/p>\n<p>of patients become wheelchair bound within 15 years of disease onset and<\/p>\n<p>during the last stages of the disease, patients are bedridden. According<\/p>\n<p>to the National Multiple Sclerosis Society, an estimated 1,600 to 1,800<\/p>\n<p>Nebraskans have MS.<\/p>\n<p>The cause of MS remains elusive, but most people have a normal life<\/p>\n<p>expectancy. The vast majority of MS patients are mildly affected, but in<\/p>\n<p>the worst cases, MS can render a person unable to write, speak, or walk.<\/p>\n<p>MS patients require lifelong medical care and rehabilitation.
