A University of Nebraska<\/p>\n<p>Medical Center biochemist, Dr. Sanderson was conducting some experiments<\/p>\n<p>several years ago to determine how certain tiny molecules called peptides<\/p>\n<p>interact with the immune system. One peptide surprisingly jumped out at<\/p>\n<p>him in the experiments.<\/p>\n<p>It caught my attention right away, said Dr. Sanderson, who is associate<\/p>\n<p>professor in the UNMC School of Allied Health Professions. I was intrigued<\/p>\n<p>by how it interacted with cell receptors. I knew I was on to something.<\/p>\n<p>After years of research, that something turned out to be a possible<\/p>\n<p>vaccine that would eliminate a smokers craving for nicotine. Earlier this<\/p>\n<p>month, Dr. Sandersons efforts were rewarded with a one-year, $100,000<\/p>\n<p>Small Business Technology Transfer Research (STTR) grant from the National<\/p>\n<p>Institute of Drug Abuse.<\/p>\n<p>The grant was awarded to Dr. Sandersons own company called Prommune,<\/p>\n<p>a spin-off company that was developed through UNeMed, a company created<\/p>\n<p>by UNMC for the purpose of getting UNMC research breakthroughs into the<\/p>\n<p>marketplace.<\/p>\n<p>The grant is a partnership between a for-profit company (Prommune) and<\/p>\n<p>a non-profit research institution (UNMC). The whole idea behind the grant<\/p>\n<p>is to bring these two entities together so that promising research discoveries<\/p>\n<p>can be put on the fast track, Dr. Sanderson said. The hope is that the<\/p>\n<p>partnership will allow these products to get into the marketplace sooner<\/p>\n<p>and into the hands of people who can benefit from them.<\/p>\n<p>Using the serendipitous peptide that Dr. Sanderson discovered, a simple<\/p>\n<p>vaccine can be created that sensitizes the immune system to recognize nicotine<\/p>\n<p>as a foreign molecule. The idea is that when you smoke a cigarette, your<\/p>\n<p>immune system generates nicotine-specific antibodies, which bind up the<\/p>\n<p>nicotine in peripheral circulation, thus preventing the nicotine from getting<\/p>\n<p>into the brain where it induces its pleasurable and addictive effects,<\/p>\n<p>Dr. Sanderson said.<\/p>\n<p>The ability of Dr. Sandersons nicotine vaccine to reduce nicotine addiction<\/p>\n<p>was demonstrated in experimental rats. These results were recently published<\/p>\n<p>in the March 2003 issue of the scientific journal, International Immunopharmacology.<\/p>\n<p>We measured some of the behavioral traits of the rats, such as how they<\/p>\n<p>moved around when they were given nicotine, Dr. Sanderson said. We were<\/p>\n<p>able to prove that the vaccine really did negate the typical behavior that<\/p>\n<p>occurred when the rats were given nicotine.<\/p>\n<p>Under Phase I of the STTR grant, Dr. Sanderson will have one year to<\/p>\n<p>prove that the nicotine vaccine is compatible with humans. If he can accomplish<\/p>\n<p>this, he can then qualify for a Phase II STTR grant worth $750,000. Under<\/p>\n<p>Phase II, he would apply for an Investigational New Drug (IND) with the<\/p>\n<p>intent of determining the vaccines efficacy in humans. If all goes well,<\/p>\n<p>Dr. Sanderson hopes the vaccine will be available for clinical trials in<\/p>\n<p>humans within a couple years.<\/p>\n<p>The beauty of this vaccine is that its completely synthetic, Dr.<\/p>\n<p>Sanderson said. We can make huge amounts of it in about three or four<\/p>\n<p>days. It is produced as a dry white powder and has a shelf life of several<\/p>\n<p>years.&nbsp; It needs no refrigeration and vaccination is accomplished<\/p>\n<p>by a small injection of this dry powder dissolved merely in water.&nbsp;<\/p>\n<p>The production of this vaccine is very straight forward, and it will allow<\/p>\n<p>us to move forward with considerable dispatch in making it available to<\/p>\n<p>those who really need it.<\/p>\n<p>Dr. Sanderson is hopeful that the vaccine would be delivered by a patch<\/p>\n<p> not by needle injection. The vaccine targets the Langerhans cells, which<\/p>\n<p>are antigen-presenting cells located just below the top layer of skin.<\/p>\n<p>Dr. Sanderson said the nicotine vaccine could be used as a platform<\/p>\n<p>for developing vaccines for other addictive drugs, such as methamphetamine.<\/p>\n<p>In addition, the Department of Defense feels the vaccine has the potential<\/p>\n<p>of being used to help develop protection against agents of terrorism, such<\/p>\n<p>as nerve gases, anthrax and tularemia.<\/p>\n<p>Its very exciting, but we still have a lot of work to do, Dr. Sanderson<\/p>\n<p>said. The cigarette addiction is extremely powerful. The nicotine vaccine<\/p>\n<p>wont suffice alone in getting people to quit smoking, but by taking away<\/p>\n<p>the craving for nicotine, it would be an important step in getting people<\/p>\n<p>to quit. I look at it as just another tool to help people get the smoking<\/p>\n<p>monkey off their backs.<\/p>\n<p>Dr. Sanderson said that on the basis of scores he and his team have<\/p>\n<p>received, he expects funding of a second grant that would involve another<\/p>\n<p>aspect of research on the nicotine vaccine. This grant would be a three-year,<\/p>\n<p>$900,000 grant from the National Cancer Institute and would start July<\/p>\n<p>1.<\/p>\n<p>The nicotine vaccine research team also includes Jonathan Vennerstrom,<\/p>\n<p>Ph.D., associate professor in the UNMC Department of Pharmaceutical Science,<\/p>\n<p>Geoffrey Thiele, Ph.D., professor in the UNMC Department of Internal Medicine,<\/p>\n<p>and Rick Bevins, Ph.D., associate professor in the Department of Psychology<\/p>\n<p>at the University of Nebraska-Lincoln.<\/p>\n<p>-30-<\/p>\n<p>&nbsp;<\/p>\n<\/div><\/div>","protected":false},"excerpt":{"rendered":"<p>Sam Sanderson, Ph.D., knows all about serendipity.
