{"id":93553,"date":"2022-11-16T15:25:00","date_gmt":"2022-11-16T21:25:00","guid":{"rendered":"https:\/\/www.unmc.edu\/newsroom\/?p=93553"},"modified":"2022-11-16T15:21:55","modified_gmt":"2022-11-16T21:21:55","slug":"unmc-researcher-helps-create-new-tool-for-crispr-toolbox","status":"publish","type":"post","link":"https:\/\/www.unmc.edu\/newsroom\/2022\/11\/16\/unmc-researcher-helps-create-new-tool-for-crispr-toolbox\/","title":{"rendered":"UNMC researcher helps create new tool for CRISPR toolbox"},"content":{"rendered":"<div class=\"panel body-content\"><div class=\"panel__container\">\n<p>UNMC researcher Channabasavaiah Gurumurthy, PhD, MVSC (DVM), contributing to a team from Tokai University, Japan, has co-authored a <a href=\"https:\/\/genomebiology.biomedcentral.com\/articles\/10.1186\/s13059-022-02779-8\" target=\"_blank\" rel=\"noreferrer noopener\">recent publication in the journal Genome Biology<\/a> that heralds an exciting new advance in the use of CRISPR technology.<\/p>\n\n\n\n<p>The discovery could become a key contribution toward the use of gene editing in many research fields, leading to developing human therapeutics.<\/p>\n\n\n\n<p>CRISPR, which gave scientists the ability to \u201ccut\u201d individual genes from a genome, revolutionized the research world and won a Nobel Prize.<\/p>\n\n\n\n<p>Then, Easi (Efficient additions with ssDNA inserts)-CRISPR, for which <a href=\"https:\/\/www.unmc.edu\/newsroom\/2017\/05\/18\/easi-crispr-a-breakthrough-in-genome-engineering\/\" target=\"_blank\" rel=\"noreferrer noopener\">Dr. Gurumurthy was a primary investigator<\/a>, made the process easy. And <a href=\"https:\/\/www.unmc.edu\/newsroom\/2018\/03\/01\/companion-technology-makes-easi-crispr-easier\/\">then easier<\/a>.<\/p>\n\n\n\n<p>\u201cCRISPR is a scissors that cuts genes,\u201d Dr. Gurumurthy said. \u201cSo in the original CRISPR, it is search, find and cut. But in our case Easi-CRISPR is search, find, cut and insert.\u201d<\/p>\n\n\n\n<p>This allows researchers to generate custom genetically engineered (knock-in) animal models at previously unheard-of rates and significantly lower cost.<\/p>\n\n\n\n<p>When Easi-CRISPR came out in 2017, Paula Turpen, PhD, now-retired director of research resources at UNMC, likened the jump in tech to going from leafing through pages to make corrections with \u201cWite-Out\u201d on a manual typewriter to simply using Microsoft Word\u2019s \u201cfind and replace\u201d feature.<\/p>\n\n\n\n<p>It was an incredible advance for basic research.<\/p>\n\n\n\n<p>But living cells that help with the \u201cinsert\u201d step are unpredictable. A cut here could cause another change there. And blanket \u201cfind and replace\u201d commands sometimes lead to what Dr. Gurumurthy calls \u201coff-target editing events.\u201d Essentially, that is the unintended consequence of a piece of DNA inserting itself not just where the researcher sent it, but also somewhere else.<\/p>\n\n\n\n<p>\u201cEven though (Easi-CRISPR) is highly accurate, there is a slight chance that there can be missed perfections,\u201d Dr. Gurumurthy said.<\/p>\n\n\n\n<p>Now, \u201cthose missed perfections can be identified using the CRISPR-KRISPR method,\u201d Dr. Gurumurthy said.<\/p>\n\n\n\n<p>CRISPR-KRISPR: CRISPR-Knock-ins and Random Inserts Searching PRotocol. As the full name indicates, CRISPR-KRISPR searches for those random inserts, allowing scientists to double-check their work.<\/p>\n\n\n\n<p>Currently, CRISPR-edited genomes that carry this risk are largely used in human trials in cases in which patients are terminally ill and there are few other treatment options.<\/p>\n\n\n\n<p>But CRISPR-KRISPR scans for \u201cmissed perfections\u201d to remove them before the edited genome is inserted. And that could open a world of possibilities for human therapeutics.<\/p>\n\n\n\n<p>Dr. Gurumurthy is encouraged that the <a href=\"https:\/\/genomebiology.biomedcentral.com\/articles\/10.1186\/s13059-022-02779-8\" target=\"_blank\" rel=\"noreferrer noopener\">journal article<\/a>, already accessed over 2,500 times in just a few weeks, and his <a href=\"https:\/\/twitter.com\/CRISPRGuru\/status\/1587899682511982592\" target=\"_blank\" rel=\"noreferrer noopener\">explanatory Twitter thread<\/a> on CRISPR-KRISPR have built buzz in the research world.<\/p>\n\n\n\n<p>\u201cEasi-CRISPR caught on within a couple months, and we are thinking it will be similar to that,\u201d he
