Notes on Sample Types:
To save money, we recommend that you premix your samples and submit them as Premixed.
A Predefined sample is submitted at a higher DNA concentration than required and in a tube separate from the primer. The primer should be in a separate tube at 5uM; include at least 5uL of primer for each sequencing reaction you request for that primer. You must measure the sample DNA concentration and provide that information on your order form. You must include enough DNA and primer to equal or exceed the requirements for the number of reactions you request. Genewiz will dilute your DNA sample for you, and use the appropriate amount of DNA and primer in their process.
A Custom sample is submitted at a higher DNA concentration in a tube separate from the primer. You must include enough DNA and primer to equal or exceed the requirements for the number of sequencing reactions request for them. The primer must be at a concentration of 5uM or higher. Thus, a Custom sample is a sample whose DNA concentration you want Genewiz to measure for you and optimize the amount for the sequencing work.
A difficult sequence sample is DNA that is GC rich, a hairpin, RNAi sequence or other difficult to sequence DNA.
